The fifth base in human DNA, 5‐methylcytosine, is inherently mutagenic. This has led to marked changes in the distribution of the CpG methyl acceptor site and an 80% depletion in its frequency of occurrence in vertebrate DNA. The coding regions of many genes contain CpGs which are methylated in sperm and serve as hot spots for mutation in human genetic diseases. Fully 30–40% of all human germline point mutations are thought to be methylation induced even though the CpG dinucleotide is (...) under‐represented and efficient cellular repair systems exist. Importantly, tumor suppressor genes such as p53 also contain methylated CpGs and these serve as hot spots for mutations in some, but not all, human cancers. Comparison of the spectrum of mutations present in this gene in different human cancers allows for predictions to be made on the molecular mechanisms of tumorigenesis. (shrink)

The ‘What Being the Parent of a New Baby is Like-Revised’ is an instrument designed to measure adaptation to parenthood. In the process of pilot testing and validating the WPL-R in a postpartum Dutch population, we became aware of the potentially sensitive nature of the measure. Despite the ethics committee waiving the invasive nature of the measure, we conducted a survey to explore its possible effect on women’s thoughts and emotions by using the Positive And Negative Affect Schedule to measure (...) changes associated with completion of the WPL-R. Two hundred and fifteen questionnaires were returned. Our findings indicated a change in PANAS scores, implying an intervening effect when using the WPL-R, in research. This raises the question of whether it is acceptable to use a questionnaire with postpartum women without any knowledge of the possible effect it may have, even though the ethics committee has approved the study and considers the measure to be non-invasive. Women relied on both justice as well as utilitarianism when making moral decisions. By contrast, men relied only on justice, and did not rely on utilitarianism (...) when faced with the same ethical issues. Further, women's intention to behave was contextual, significantly affected by two national culture dimensions (uncertainty avoidance and individualism), whereas men's decisions were more universal, and not related to national culture dimensions. In this comparative survey of 126 Brazilian and U.S. business professionals, we explore the effect of national culture on ethical decision-making within the context of business. Using Reidenbach and Robin''s (1988) multi-criteria ethics instrument, we examined how these two countries'' differences on Hofstede''s individualism/collectivism dimension are related to the manner in which business practitioners make ethical decisions. Our results indicate that Brazilians and Americans evaluate the ethical content of actions or decisions differently when applying utilitarian criteria. By contrast, business people (...) from both countries do not differ significantly when they use egoistic criteria in evaluating the ethical nature of business decisions. (shrink)

BackgroundBiobanks are considered to be key infrastructures for research development and have generated a lot of debate about their ethical, legal and social implications. While the focus has been on human genomic research, rapid advances in human microbiome research further complicate the debate.DiscussionWe draw on two cystic fibrosis biobanks in Toronto, Canada, to illustrate our points. The biobanks have been established to facilitate sample and data sharing for research into the link between disease progression and microbial dynamics in the lungs (...) of pediatric and adult patients. We begin by providing an overview of some of the ELSI associated with human microbiome research, particularly on the implications for the broader society. We then discuss ethical considerations regarding the identifiability of samples biobanked for human microbiome research, and examine the issue of return of results and incidental findings. We argue that, for the purposes of research ethics oversight, human microbiome research samples should be treated with the same privacy considerations as human tissues samples. We also suggest that returning individual microbiome-related findings could provide a powerful clinical tool for care management, but highlight the need for a more grounded understanding of contextual factors that may be unique to human microbiome research.ConclusionsWe revisit the ELSI of biobanking and consider the impact that human microbiome research might have. Our discussion focuses on identifiability of human microbiome research samples, and return of research results and incidental findings for clinical management. (shrink)

What role does the wild duck play in Ibsen's famous drama? I argue that, besides mirroring the fate of the human cast members, the duck is acting as animal subject in a quasi-experiment, conducted in a private setting. Analysed from this perspective, the play allows us to discern the epistemological and ethical dimensions of the new scientific animal practice (systematic observation of animal behaviour under artificial conditions) emerging precesely at that time. Ibsen's play stages the clash between a scientific and (...) a romantic understanding of animals that still constitutes the backdrop of most contemporary debates over animals in research. Whereas the scientific understanding reduces the animal's behaviour, as well as its environment, to discrete and modifiable elements, the romantic view regards animals as being at one with (or violently disconnected from) their natural surroundings. In this paper, I will reread the history of molecular genetics from a psychoanalytical angle, analysing it as a case history. Building on the developmental theories of Freud and his followers, I will distinguish four stages, namely: (1) oedipal childhood, notably the epoch of model building (1943–1953); (2) the latency period, with a focus on the development of basic skills (1953–1989); (3) adolescence, exemplified by the Human Genome Project, with its fierce conflicts, great expectations and grandiose claims (1989–2003) and (4) (...) adulthood (2003–present) during which revolutionary research areas such as molecular biology and genomics have achieved a certain level of normalcy—have evolved into a normal science. I will indicate how a psychoanalytical assessment conducted in this manner may help us to interpret and address some of the key normative issues that have been raised with regard to molecular genetics over the years, such as ‘relevance’, ‘responsible innovation’ and ‘promise management’. Probably to most students of Moral Philosophy there comes a time when they feel a vague sense of dissatisfaction with the whole subject. And the sense of dissatisfaction tends to grow rather than to diminish. It is not so much that the positions, and still more the arguments, of particular thinkers seem unconvincing, though this is true. It is rather that the aim of the subject becomes increasingly obscure. "What," it is asked, "are we really going to learn by Moral (...) Philosophy?" "What are books on Moral Philosophy really trying to show, and when their aim is clear, why are they so unconvincing and artificial?" And again: "Why is it so difficult to substitute anything better?" Personally, I have been led by growing dissatisfaction of this kind to wonder whether the reason may not be that the subject, at any rate as usually understood, consists in the attempt to answer an improper question. And in this article, I shall venture to contend that the existence of the whole subject, as usually understood, rests on a mistake, and on a mistake parallel to that on which rests, as I think, the subject usually called the Theory of Knowledge. THIS paper seeks to elucidate the phenomenon known in psychology as 'the specious present,' by postulating a two-dimensional theory of the extensional aspects of time. On this theory, the usual logical and psychological difficulties, encountered in current accounts of this phenomenon, can be resolved. For, when there are two dimensions of time, the same event may be without extension in one of these dimensions ('transition-time'), while it is nevertheless finitely extended in the other of these dimensions ('phase-time'); so that in (...) a definable sense the phases of a finitely enduring event, though successive in one time-order, yet are contemporary in the other. The epistemological standpoint implicit in the paper is generally similar to the one Bertrand Russell has put forward, in his Physics and Experience (Cambridge, 1946), and his Human Knowledge: Its Scope and Limits (London, 1948) (allowing for the changes which would be required in Russell's theory to take into account a second time dimension). A 'psycho-neural parallelism,' or one-one correspondence, is postulated between features of certain 'experiential events' (namely, those experiential events normally held to be happening to some person's mind, which are describable in the language of psychology); and features of certain 'physical events' (namely, those events described in the language of physics, chemistry and physiology, which are ordinarily conceived as happenings in that same person's body). These physical events are conceived of as being causally connected with events in the physical world outside the experient's body, by means of the concepts of light waves, sound waves, chemical stimuli, and consequential processes in the nervous system (central and peripheral) and sense-organs, in the usual way. In terms of this psycho-neural parallelism the physical correlate of the finite temporal span of the specious, or experiential present, is to be found in certain consequences of the uncertainty principle in quantum physics; according to which there is a finite interval of time necessarily associated with a nearly precise determination of energy levels, and of transitions between them. Some of the physical implications of this theory, applied to processes in the material structure of the human body, are discussed qualitatively. But, for the reasons given in the last section of the paper, a quantitative treatment is not yet possible. The paper is greatly indebted in regard to the physical application of the two-dimensional theory of time, to the discussion of the pentadic group structure in Eddington's Fundamental Theory (Cambridge, 1946 and 1950); and in particular to the treatment there of the phase-variable as the 'time-analogue' in the quantum statistics of stationary states. Populist attributions of blame have important effects on citizens’ attitudes, cognitions, emotions, and behaviors. Extending previous studies that have mostly looked at populist messages blaming political elites, we use an online survey experiment (N = 805) to investigate the effects of blaming different elitist actors in populist and non-populist ways: (1) political elites, (2) corporate elites, (3) scientific elites, and (4) a combination of these elites. We compare mere causal responsibility attribution to populist blame attributions that highlight a central opposition (...) between “pure ordinary people” and “corrupt elites” to tease out the specific effect of populist rhetoric. Results suggest that populist messages blaming all elites have stronger effects than isolated cues. Furthermore, populist blame attribution leads to more negative perceptions of the elites than non-populist blame attribution. The finding that populist frames can – under some conditions – activate negative perceptions toward different elites is alarming in times of declining trust in societal institutions, and calls for a more encompassing understanding of populists’ scapegoats.
